UV-B辐射对小麦根茎叶中钙调蛋白含量的影响 被引量:1

摘要 以荧光图谱法探讨增强UV-B辐射对钙调蛋白的影响。以8.00kJ/(m2.d)、10.08kJ/(m2.d)、12.00kJ/(m2.d)3种不同剂量UV-B辐射的小麦为材料,经过多次离心、(NH4)2SO4盐析沉淀法、透析法、疏水层析等步骤得到了钙调蛋白的提取液,通过SDS-PAGE测定钙调蛋白的相对分子质量,并检验纯度。结果显示:分离纯化的钙调蛋白分子量为16 000左右,电泳图谱为一条带。用荧光分光光度计绘制钙调蛋白的荧光图谱,同时设置多组对照试验。结果显示:根茎叶中钙调蛋白的含量不同,而且增强UV-B辐射后小麦中整体钙调蛋白含量呈现增加趋势,根茎叶中钙调蛋白受到不同的UV-B处理后变化程度不同,茎中钙调蛋白随着UV-B辐射的影响变化最为明显,根中钙调蛋白的变化次之,叶片中钙调蛋白变化最少。 This article explored effects of enhanced UV-B radiation on calmodulin(CaM),by using three different doses of UV-B radiation,i.e.8.00kJ/(m2·d),10.08kJ/(m2·d) and 12.00kJ/(m2·d).The results showed that the calmodulin purified had a molecular weight of about 16 000.The CaM contents in root,stem and leaf were different.After strengthening the UV-B radiation,the calmodulin content increased in wheat,with varying degree in different tissues.The change of the CaM content in stem was the greatest and that in leaf was the least.
