Relationship between ghrelin gene Leu72Met polymorphism and type 2 diabetes mellitus

摘要 目的探讨Ghrelin基因Leu72Met多态性与T2DM之间的相关性。方法采集316例T2DM患者和203名健康对照组(NGT组)人群的一般临床资料,利用PCR-RFLP进行Ghrelin基因Leu72Met的多态性分析。结果 T2DM组的Leu72Met基因型分布及等位基因频率与NGT组相比差异无统计学意义。NGT Met72携带者血清LDL-C水平较同组Leu72纯合子个体增高(P<0.05)。结论 Ghrelin基因Leu72Met分布与等位基因频率在T2DM和健康人群无明显差异,该基因多态性在健康人群可能与血清LDL-C水平相关。 Objective To investigate the relationship between ghrelin Leu72Met polymorphisms and type 2 diabetes. Methods 316 type 2 diabetic patients and 203 health people were genotyped for Leu72Met of ghrelin by PCR-RFLP assay. Anthropometric variables, blood glucose, insulin, lipids and other clinical parameters were measured. Results In control group, the genotype frequencies were as follows: CC 88. 2% (n= 179), CA 10. 3% (n=21), and AA 1.5% (n=3). In type 2 diabetes group, the genotype frequencies were as follows: CC86. 1% (n=272), CA 11.7% (n=37), and AA 2.2% (n=7). There were no significant differences in the genotype frequencies between control and T2DM groups. Serum LDL-C was higher in subjects carrying Met72+ genotype than in subjects with wild genotype in control group (P〈0. 05). Conclusion The allele and genotype fregueneies distribution of the Leu72Met is not different between control and type 2 diabetes groups Leu72Met variants is related with serum LDL-C in control group.
