摘要
为了提高羊肚菌胞外多糖得率,对羊肚菌液体培养进行了研究。通过单因素实验确定了最优碳源为玉米粉,最优氮源为蛋白胨,最佳生长因子为麸皮;采用正交实验,确定了羊肚菌发酵的最优培养基配方为:玉米粉3%,蛋白胨0.3%,麸皮2%,MgSO40.2%,KH2PO40.2%;并采用二次回归正交组合设计确定了最优培养条件为:温度25℃,摇床转速96r/min。在发酵罐扩大实验中,对胞外多糖含量进行了监测,其中发酵108h时胞外多糖含量达1.145g/L。
This article researched on submerged fermentation of Morchella to improve the exopolysaccharides yield.By the method of single-factor test,corn flour,peptone,wheat bran were choosed as the optimum carbon source,nitrogen source and growth factor respectively.By carrying on the orthogonal experiment,the optimum fermentation medium was determined as follows:corn flour 3%,peptone 0.3%,wheat bran 2%,MgSO4 0.2%,KH2PO4 0.2%.The result of the orthogonal regression design indicated that the optimum culture temperature was 25 ℃ and frequency of shake-flask was 96 r/min.The content of exopolysaccharides was monitored in the fermentor,and it could reach 1.145 g/L after 108 h.
