摘要
为建立一种简便、快捷、灵敏的HGAG的测定方法,研究了天青Ⅰ染色剂直接测定HGAG含量的分光光度法,二者能特异性结合成蓝紫色复合物,该复合物在特定波长处有最大光吸收,通过测定吸光度值可测HGAG含量。结果天青Ⅰ-HGAG复合物在波长547nm处有最大光吸收,且在5~40mg/L范围内,具有良好的正相关线性关系,回归方程是A=0.0087x+0.2057,R2=0.9914;该测定方法在3h内稳定性、重现性测定结果P>0.05,说明数据可靠,加样平均回收率达95.47%,是一种简便、快捷、灵敏的测定方法。
Aimed to establishing a simple,rapid and sensitive method to determine the content of holothurian glycosaminoglycan,the research study on directly spectrophotometric determination about the content of holothurian glycosaminoglycan with AzureⅠ.They could form a specific-binding blue-purple complex that was Azure Ⅰ-holothurian glycosaminoglycan.At the characteristic wavelength,this complex had the max absorption,and the content of holothurian glycosaminoglycan could be determined by absorption value.The result was that the complex had the max absorption at 547 nm.A good positive linear relationship was shown in the mass concentration range of chondronitin sulfate 5~40 mg/L.The regression equation was A=0.0087x+0.2057,R2=0.9914.With this method,the result of the stability in 3 h and the repeatability was well and reliable that P﹥0.05.The recovery average rate of added samples was 95.47%.It was a simple,rapid and sensitive determination method.
