摘要
目的探讨橙皮苷对中波紫外线(UVB)诱导的小鼠表皮细胞光产物的清除作用。方法将50只小鼠随机分为对照组(A组,5只)、橙皮苷1 mg/cm2组(B组,5只)、UVB 180 mJ/cm2组(C组,20只)和橙皮苷1 mg/cm2+UVB 180 mJ/cm2组(D组,20只)。采用免疫组化法及斑点免疫印迹法分别检测UVB辐射后0、24、48和72 h小鼠表皮内环丁烷嘧啶二聚体(CPDs)的产生和清除情况。结果 A组和B组小鼠表皮无明显CPDs表达;而辐射后24 h和48 h,D组CPDs表达明显低于C组(P<0.05)。结论橙皮苷能加速UVB辐射后光产物的清除,是一种有效的紫外线防护剂。
Objective To investigate the effect of hesperidin on the photoproducts induced by ultraviolet B(UVB) irradiation on the epidermal cells of mouse.Methods Fifty mice were randomly divided into four groups of A(normal controls,5 mice),B(hesperidin 1 mg/cm2,5 mice),C(UVB 180 mJ/cm2,20 mice) and D(hesperidin 1 mg/cm2 plus UVB 180 mJ/cm2,20 mice).The formation and scavenging of cyclobutane pyrimidine dimers(CPDs) at 0,24,48 and 72 h after UVB irradiation were detected by immunohistochemistry and dot Western blot.Results There was no obvious CPDs expression observed in groups of A and B.Whereas at 24 and 48 h after irradiation,the expression of CPDs in group D was significantly lower than that in group C(P〈0.05).Conclusion Hesperidin can promote the scavenging of photoproducts after irradiation and is a promising protective substance against UVB irradiation.
