地塞米松对野百合碱诱导肺动脉高压模型大鼠血管活性物质的影响

Effects of dexamethasone on the vasoactive substances in rats with monocrotaline-induced pulmonary hypertension

摘要 目的探讨地塞米松早期干预对野百合碱(MCT)诱导肺动脉高压(PAH)大鼠肺血管活性物质的影响。方法成年雄性SD大鼠30只随机均分为对照(C)组、MCT模型(M)组、MCT+地塞米松(MD)组。建立MCT诱导PAH模型,应用放射免疫法观察血浆一氧化氮(NO)和内皮素1(ET-1)水平的变化。结果注射MCT 3周后,M组大鼠肺动脉平均压(mPAP)显著高于C组和MD组(P<0.05);与C组和MD组比较,M组血浆ET-1含量升高,NO含量降低(P<0.05);而MD组ET-l和NO含量与C组比较无统计学差异。结论地塞米松能抑制MCT诱导的PAH大鼠血浆中的ET-1含量升高和NO含量降低,从而改善内皮细胞功能,降低PAH。 Objective To investigate the effects of dexamethasone on the vasoactive substances in rats with monocrotaline(MCT)-induced pulmonary arterial hypertension(PAH).Methods Thirty male SD rats were randomly divided into three groups of C(control),M(MCT-induced PAH model) and MD(PAH model treated with dexamethasone) with 10 rats each.The MCT-induced PAH model was established,and the plasma levels of nitrus oxide(NO) and endothelin-1(ET-1) were detected by radioimmunoassay.Results At 3 weeks after PAH model established,the mean pulmonary arterial pressure(mPAP) was significantly higher in group M than that in groups of C and MD(P〈0.05).The plasma ET-1 was higher,but NO was lower,in group M than those in groups of C and MD(P〈0.05),which in group MD were not significantly different from those in group C.ConclusionDexamethasone can attenuate the increase of ET-1 and decrease of NO,which improves endothelial function and decreases MCT-induced PAH in rats.
