摘要
[目的]研究蛛网膜下腔出血(SAH)大鼠突触蛋白SynapsinⅠ及其磷酸化水平的动态变化,探讨SynapsinⅠ及其磷酸化在SAH发生发展过程中的作用。[方法]将Wistar大鼠随机分入正常对照组、假手术组和SAH 1 h,1、3、5、14 d组。用枕大池二次注血法制作SAH模型,用激光多普勒血流计检测皮层局部脑血流量(r CBF)动态变化;采用免疫荧光化学及Western blot法检测皮层SynapsinⅠ及其磷酸化蛋白表达。[结果]SAH后r CBF动态下降,1 h、3 d组下降明显(P<0.01)。SAH各亚组顶叶皮层SynapsinⅠ表达均下降,1、3、5 d组差异有显著性(P<0.01),磷酸化SynapsinⅠ表达持续低水平,3 d组最明显(P<0.01)。[结论]SAH后存在突触受损及突触传递障碍。
[Objective]This study aimed to investigate the possible role of phosphorylation of synapsin I in the process of cerebral injury following subarachnoid hemorrhage(SAH). [Methods]Wistar rats were divided into non-SAH,sham-operated group,SAH60min,SAH1d,SAH3d,SAH5d and SAH 14d groups.double injections of blood into cistern magna of rats were performed to induce SAH.Synapsin I protein and phosphosynapsin I protein were measured from cerebral cortex by immunohistochemistry and Western blot. [Results]Synapsin I and phosphosynapsin I positive cells were found in frontal penum-bral cortex in every group.The expression of synapsin I was significantly decreased in the SAH1d,3 d and 5 d groups(P0.01).Expression of phosphosynapsin I was also significantly decreased.A considerable reduction of phosphosynapsin I was observed in SAH 3 d groups(P0.001),suggesting a selective defect in synapsin I phosphorylation. [Conclusion]It is concluded that phosphorylation of presynaptic proteins is probably impaired before cerebral vasospasm following SAH.The synaptic transmission may be blocked in cerebral injury following SAH.
