母胎界面MCP-1表达异常与不明原因早期复发性流产的关系

Abnormal expression of monocyte chemotactic protein-1 in pathogenesis of unexplained early recurrent spontaneous abortion

目的:探讨绒毛、蜕膜组织中单核细胞趋化蛋白-1(MCP-1)表达异常与不明原因早期复发性流产(UERSA)的关系。方法:选取UERSA患者30例(UERSA组)以及同孕龄正常早孕妇女30例(正常早孕组),应用实时荧光定量聚合酶链反应(RT-PCR)技术检测两组绒毛、蜕膜组织中MCP-1mRNA的表达水平,应用酶联免疫吸附测定(ELISA)方法检测两组血清中MCP-1蛋白的含量。结果:早孕绒毛、蜕膜组织均有MCP-1mRNA的表达;UERSA组绒毛、蜕膜组织中MCP-1mRNA的表达量(1.96±0.39,3.67±0.98)均较正常早孕组(1.42±0.28,1.90±0.85)显著升高(P<0.01);UERSA组血清MCP-1蛋白含量为85.74±12.31pg/ml,较正常早孕组(59.41±9.70pg/ml)显著升高(P<0.01);血清MCP-1蛋白的含量与绒毛、蜕膜组织中MCP-1mRNA的水平分别呈正相关(r=0.595,0.655)。结论MCP-1表达于早孕母胎界面,绒毛、蜕膜组织MCP-1mRNA高表达可能与UERSA的发病机制有关,提示血清MCP-1升高在胚胎停育的早期诊断中具有重要的临床意义。

To investigate the relationship between abnormal expressions of monocyte chemotactic protein - 1 （ MCP -1 ） mRNA in villus and decidua and unexplained early recurrent spontaneous abortion （UERSA）. Methods： The expres- sions of MCP -1 mRNA in villus and decidua tissues of 30 UERSA patients （UERSA group） and 30 normal early pregnancy women （normal early pregnancy group） were determined by real - time fluorescence quantitative PCR. The maternal serum levels of MCP - 1 were detected by ELISA. Results ： The MCP - 1 mRNA expressions in first - trimester placental villus and decidua tissues in UERSA group were 1.96 ＋ 0.39 and 3.67 ＋ 0.98 respectively, which were higher than those in normal early pregnancy group （1.42±0.28, 1.90±0.85, P 〈0.01 ）. The concentration of serum MCP - 1 in UERSA group was 85. 74±12.31 pg/ml, higher than that in normal early pregnancy group （59.41±9.70 pg/ml, P 〈 0.01 ）. The serum MCP- 1 level was positively correlated with MCP - 1 mRNA levels in villus and decidua （ r = 0. 595 and 0. 655 respectively, P 〈 0. 01 ）. Conclusion ： MCP - 1 was expressed in villus and deciduas. The higher MCP - 1 mRNA expression might play an impor- tant role in the pathogenesis of UERSA. The elevated concentration of serum MCP - 1 could be used as a tool for diagnosing embryonic death.
