摘要
目的检测KISS-1蛋白、基质金属蛋白酶(MMP)-9及Ki67在前列腺癌中的表达,探讨三者在前列腺癌发生发展过程中的作用。方法采用免疫组织化学法检测15名正常前列腺组织、20例前列腺上皮内瘤(PIN)、48例前列腺癌组织KISS-1蛋白、MMP-9及Ki67的表达,分析三者在不同临床病理特征中表达的意义及它们之间的相关性。结果前列腺癌组KISS-1蛋白阳性表达率(42%)明显低于正常前列腺组织和前列腺PIN组(分别为93%、75%;P<0.01),并且KISS-1蛋白低表达与前列腺癌的临床分期和淋巴结转移有关(P<0.05),与前列腺癌患者的年龄、组织学分级无关(P>0.05)。前列腺癌组和前列腺PIN组MMP-9阳性表达率(分别为79%,55%)明显高于前列腺组(13%;P<0.01),并且MMP-9高表达与前列腺的临床分期和淋巴结转移有关(P<0.05),与前列腺癌患者的年龄、组织学分级无关(P>0.05)。前列腺癌组和PIN组的Ki67阳性表达率(分别为85%,60%)明显高于正常前列腺组(13%;P<0.01),并且Ki67高表达与前列腺癌的临床分期有关(P<0.05),与前列腺癌患者的年龄、组织学分级和淋巴结转移无关(P>0.05)。在前列腺癌中KISS-1蛋白与MMP-9表达呈负相关(r=-0.399,P<0.01)。结论 KISS-1蛋白、MMP-9及Ki67与前列腺癌的发生发展有关,联合检测三者的表达可能作为评估前列腺癌预后和指导临床靶向治疗的重要指标。
Objective To investigate the expression of KISS-1 protein, matrix metalloprotease (MMP)-9 and Ki67, and their relationship in squamous cell carcinoma of the prostate. Methods The expression of KISS-1 protein, MMP-9 and Ki67 was examined by immunohistochemical assay (SP) in 15 cases of normal prostate epilhelia, 20 cases of prostatic intraepithelial neoplasia (PIN), 48 cases of prostatic cancer (PCa), so as to analyze the relationship between them and the characteristics of clinical pathology of PCa. Results It was found that the positive rate of KISS-I protein expression in PCa group(42%) was significantly less than in PIN and normal prostate epithelial tissue group (93%, 75%, P〈0.01 ). The low expression of KISS-1 protein was correlated with clinical stage and lymph node metastasis of PCa (P〈 0.05 ), but was not correlated with age of patients and histological grade of PCa. The positive rate of MMP-9 expression in PCa and PIN group (79%, 55%) was significantly higher than that in normal prostate epithelial tissue group (13%, P〈 0.01 ). Conclusion The expression of KISS-l protein, MMP-9 and Ki67 is associated with the occurrence and development of PCa. Combined analysis of these three proteins may be an important index to judge prognosis and direct clinical targeted therapy of PCa.
