摘要
以欧李仁蛋白为底物,采用碱性蛋白酶、中性蛋白酶和酸性蛋白酶进行分步复合酶解,以水解度为指标,确定其分步复合酶解的条件。结果表明,碱性蛋白酶的最适条件为底物质量分数5%,酶添加量为1.5%(基于底物蛋白质的质量),温度50℃,pH值10;中性蛋白酶添加量为5%,温度40℃,pH值7;酸性蛋白酶添加量5%,温度50℃,pH值5。分别水解30 min,经这3种酶酶解后其多肽质量浓度可达27.566 1 mg/mL。
Using Cerasus Humilis Kernel protein as substrate,the substrate is hydrolyzed by Alcalase,Neutrase and acid proteinase in turn,the effects of substrate concentration,enzyme concentration and time of enzymatic hydrolysis on hydrolysis degree in Cerasus Humilis Kernel protein are studied in this paper.The optimal conditions of hydrolysis are obtained by experiment as follows:substrate concentration 5%,Alcalase enzyme concentration 1.5% of substrate,pH 10,temperature 50 ℃,Neutrase proteinase concentration 5%,pH 7,temperature 40 ℃,acid proteinase concentration 5%,pH 5, temperature 50 ℃.The substrate is hydrolyzed for 30 min respectively in optimum conditions of these enzymes.The concentration of peptide is up to 27.566 1 mg/mL after hydrolysis of these three enzymes.
