摘要
【目的】阐明广西中华草龟的遗传多样性,为今后保护其种质资源及基因库、防止规模化养殖后造成的基因流失提供理论指导,也为中华草龟合理开发利用及选择育种提供科学依据。【方法】运用RAPD技术从分子水平上对广西中华草龟遗传多样性进行分析。【结果】从7只广西中华草龟的基因组中共扩增出267条条带,平均每个个体约扩增出38条条带,平均每条随机引物获得15条以上的条带,扩增片段大小为100~2000bp,多态性频率为66.29%;个体间的遗传距离指数为0.1360~0.3609,平均遗传距离指数为0.2092±0.0623。【结论】广西中华草龟的遗传多样性较丰富,但个体间的亲缘关系较近,遗传变异度较小。
【Objective】The current experiment was conducted to analyze the genetic diversity amongst Chinemys reevesii accessions of Guangxi in order to protect and characterize the germplasm resources and promote its suitable development and selection breeding. 【Method】Genetic diversity of Chinemys reevesii accessions of Guangxi was analyzed using RAPD markers. 【Result】The results showed amplification of total of 267 bands from genomic DNA of 7 Chinemys reevesii accessions with an average of 38 bands per accession. Each random primer produced over 15 bands on an average. The size of fragments ranged from 100-2000 bp. Out of 267 bands, 177 were polymorphic with 66.29% polymorphism frequency. The genetic distance between individuals was 0.1360-0.3609, the average genetic distance was 0.2092±0.0623. 【Conclusion】 Abundant genetic diversity was observed amongst different accessions of Chinemys reevesii in Guangxi.
