HPLC法测定雷公藤内酯醇和内酯酮的含量

Determination of Triptolide and Triptonide in Root Extracts of Tripterygium wilfordii by HPLC

目的测定雷公藤内酯醇和内酯酮的含量。方法采用高效液相色谱法(HPLC法),色谱柱:Diamonsil C18(2)柱,流动相:乙腈-水[25∶75(10 min)→45∶55(15 min)→45∶55(20 min)],流速:1mL/min,检测波长:218 nm。结果在此色谱条件下,2种成分可以完全分离;雷公藤内酯醇、雷公藤内酯酮的线性范围为:0.5625～11.25μg/mL(r=0.9998),0.65～13.0μg/mL(r=0.9999);雷公藤内酯醇、内酯酮平均回收率为98.13%、98.58%;RSD为1.87%、1.92%。结论 HPLC法可简便、准确、快速地测定雷公藤内酯醇和内酯酮的含量。

Objective To establish a HPLC method for determination of triptolide and triptonide in root extracts of Tripterygium wilfordii.Methods The separation was performed on a column of Diamonsil C18（2） with gradient elution of acetonitrile-water [25：75（10 min）→45：55（15 min）→45：55（20 min）] as mobile phase,flow rate was 1 mL·min-1,and the detective wavelength was 218 nm.Results The two components were completely separated under the chromatographic condition,the linear range of triptolide was 0.5625-11.25 μg·mL-1（r=0.9998）,and the average recovery was 98.13%（RSD=1.87%）.The linear range of triptonide was 0.65-13.0 μg·mL-1（r=0.9999）,and the average recovery was 98.58%（RSD=1.92%）.Conclusion The method is simple,accurate,rapid and suitable for determination of triptolide and triptonide in root extracts of Tripterygium wilfordii.
