摘要
目的探讨缬草提取物8-羟基松脂醇苷对Kv1.5钾通道电流的影响。方法以转基因Kv1.5HEK293细胞为受试对象,采用膜片钳技术观察10,30μmol.L-18-羟基松脂醇苷对转基因Kv1.5HEK293细胞电流的影响。结果 8-羟基松脂醇苷浓度依赖性减少Kv1.5HEK293细胞电流,电流值从(148.3±13.0)PA/PF分别降至(109.4±4.4)PA/PF和(78.6±11.3)PA/PF,抑制率分别为28.5%和37.1%。结论 8-羟基松脂醇苷对转基因Kv1.5 HEK293细胞电流有抑制作用,且呈浓度依赖性,可以部分解释缬草抗心律失常作用机制。
Objective To study effects of 8-hydroxy resin glycosides from Valeriana officinalis L.on Kv1.5 potassium channel currents. Methods The Kv1.5 HEK293 cells currents following the administration of 8-hydroxy resin glycosides at 10 and 30 μmol·L-1 were recorded by using the whole-cell patch clamp recording technique. Results 8-hydroxy resin glycosides(10 and 30 μmol·L-1) decreased Kv1.5 HEK293 cells currents in a dose-dependent manner.The peak currents(148.3±13.0) PA/PF was reduced to(109.4±4.4) PA/PF and(78.6±11.3) PA/PF,which resulted in the suppression ratio of 28.5% and 37.1%,respectively. Conclusion 8-hydroxy resin glycosides blocks Kv1.5 HEK293 cells currents dose dependently,which may contribute to the anti-arrhythmic effects of Valeriana officinalis L..
