摘要
目的:评价参黄冲剂在延缓衰老方面的作用和临床疗效。方法:将60例符合条件的衰老患者随机分为参黄冲剂治疗组和对照治疗组,每组30例。参黄冲剂组进行常规治疗基础上加用参黄冲剂治疗,每次10 g,每日3次。对照组进行常规系统治疗,并给予甲磺酸双氢麦角毒碱片,每次1 mg,每日3次。各组均连续服药8周为1疗程,观察时间为1疗程。结果:衰老患者肾阴虚、肾阳虚2种证型P16基因的序列特征未见突变改变。参黄冲剂组与对照组在治疗前的衰老患者P16基因mRNA定量表达比较,差异无统计学意义(P>0.05),二者之间具有可比性;在治疗后两组比较,差异具有统计学意义(P<0.05)。两组治疗后衰老患者临床疗效比较,参黄冲剂组30例,显效3例,有效21例,无效6例,有效率80.0%;对照组30例,显效1例,有效12例,无效17例,有效率43.3%;经统计学处理,两组间有效率比较,差异有统计学意义(P<0.01)。结论:参黄冲剂治疗老年肾虚患者,可有效改善临床衰老的临床症状,提高日常生活能力;而且参黄冲剂可减少P16基因mRNA的表达,其机制可能是通过抑制衰老细胞的增殖而达到延缓衰老的作用,从而起到抗衰老的作用,但衰老主要证型肾阴虚、肾阳虚P16基因的序列特征未见突变改变,为进一步诠释中医辨证施治衰老的机制提供了分子生物学的依据。
Objective:To evaluate the actions and clinical efficacy of Shenhuang Granule in delaying aging.Methods:60 aging patients who met the criteria were randomized into the treatment group and the control groups with 30 cases for each group.Shenhuang Granule group was given Shenhuang Granule for treatment based on the conventional treatment(agreed prescription of Jiangsu Provincial Hospital of Traditional Chinese Medicine),10 g each time,3 times a day.The control group was given conventional treatment,and Ergot poisoning alkali mesylate hydrochlorothiazide tablets,1 mg each time,3 times a day.Each group was continuously taking 8 weeks as a course of treatment and the observation time was one course of treatment.Results:There were no mutation changes in the sequence characteristics of expressions of gene P16 in the two syndromes of aging patients,kidney-yin deficiency and kidney-yang deficiency.Quantitative expressions of mRNA in gene P16 of the aging patients in the Shenhuang Granule group and control group had no statistical significance(P0.05) with comparability between the two groups;comparison between the two groups had statistical significance after treatment(P0.05).After treatment,clinical effects were compared.Of 30 cases in the Shenhuang Granule group,3 cases were cured;21 cases were effective;6 cases were ineffective and the total effective rate was 80.0%;Of 30 patients in the control group,1 cases were cured;12 cases were effective;17 cases were ineffective and the total effective rate was 43.3%;After the statistically processing,the effective rates between the two groups were statistically significant(P0.01).Conclusion:Shenhuang Granule can be used for the treatment of elderly patients with kidney deficiency,which can effectively improve the clinical symptoms of aging and improve daily living abilities.Shenhuang Granule can reduce mRNA expressions of gene P16,the mechanism of which may probably be that the senescence is slowed down by inhibiting proliferation of aging cells,thus playing a role in anti-aging.But the sequence characteristics of gene P16 in the two syndromes of aging patients,kidney-yin deficiency and kidney-yang deficiency have no mutation changes,which provide a molecular biology basis for further interpretation of aging mechanism by using TCM syndrome differentiation to treat aging.
