摘要
目的:探讨食管癌相关基因2(ECRG2)的多态性在新疆哈萨克族食管癌组织中的基因型分布特点及其在食管癌发生中的作用及意义。方法:应用PCR-SSCP技术检测94例哈萨克族食管癌和100例哈萨克族正常对照人群血样中ECRG2外显子4处短串联重复序列(STR)多态性基因型及其分布特点。结果:新疆哈萨克族食管癌组织及正常人群血样组织中ECRG2 STR呈多态性,可分为3种类型:TCA3/TCA3,TCA4/TCA4,TCA3/TCA4,3种基因型在食管癌和对照组中构成比分别为22.3%、30.9%、46.8%和10%、56%、34%,经卡方检验,哈萨克族食管癌及正常对照组中3种基因型总构成比有显著差别(χ^2=13.589,df=2,P〈0.01);3种基因型在哈萨克族有转移的食管癌组中分布分别为47.8%、8.7%、43.5%,在无转移的哈族食管癌组中分布为14.1%、38.0%、47.9%,两组差异有统计学意义(χ^2=13.77,df=2,P〈0.01);在哈萨克族人群中,携带TCA3/TCA3基因型个体患食管癌的风险比TCA4/TCA4基因型个体高4.06倍(95%的可信区间为1.69-9.74)。结论:ECRG2基因TCA3/TCA3基因型与TCA4/TCA4基因型比较,增加了哈萨克族患食管癌的风险。
Objective: To investigate the relationships between polymorphism of Esophageal Cancer Related Gene(ECRG2) Short Tandem Repeat(STR) and susceptibility of Esophageal Squamous Cell Carcinomas in Kazakh population in Xinjiang.Methods:ECRG2 genotypes were detected by PCR-SSCP in 94 cases of Kazakh' esophageal carcinomas and 100 Kazakh' blood samples for control population.Results:The frequencies of ECRG2 STR genotypes,TCA3/TCA3,TCA4/TCA4,TCA3/TCA4 were 22.3%、30.9%、46.8% and 10%、56%、34% in Kazakh' esophageal carcinomas and the contral samples.There is a significant difference between esophageal carcinomas and the control population(χ^2=13.589,df=2,P〈0.01).The three genotypes were 47.8%、8.7%、43.5% in Kazakh' esophageal carcinomas with metastasis,and 14.1%、38.0%、47.9% in Kazakh' carcinomas without metastasis respectively.There is a significant difference between the groups with metastasis and without metastasis(χ^2=13.77,df=2,P〈0.01).Compared to those carring the TCA4/TCA4 genotype population,subjects that carried the TCA3/TCA3 genotype were at an increased risk of ESCC with the adjusted odds ratios(ORs) being 4.06(95% CI,1.69-9.74) in Kazakh.Conclusion:The results suggested that subjects those carried the TCA3/TCA3 genotype are an increased risk of ESCC and metastasis compared to those carring the TCA4/TCA4 genotype.
