摘要
目的了解在蛋白质表达水平上,肿瘤细胞减灭术和化疗对卵巢癌患者的影响,以及术前、术后及全程化疗结束后卵巢癌患者血清蛋白质组谱的变化规律。方法本实验选用同一型号的抗体芯片将健康妇女血清混合样品分别与卵巢癌患者术前、术后和化疗后血清混合样品进行杂交,分别筛选差异表达蛋白,并对各组抗体芯片结果进行比较。结果三个时间点的血清组芯片所筛选的差异表达蛋白数目,随着治疗措施的增多而减少。手术前后、化疗前后的芯片结果比较均无相同的差异表达蛋白。在三组血清芯片中共筛选出11个与细胞增殖凋亡相关的差异表达蛋白质,其数目与治疗措施呈负相关。XPA和Mcm2同步先高后低的异常表达,可能与耐药或复发有关。结论手术和化疗在卵巢癌患者的治疗中仍很重要,可能的致病因素逐渐减少。
Objective To reveal the effects of cytoreductive surgery and chemotherapy on patients with epithelial ovarian cancer (EOC) at the level of protein expression and find the regulations of protein expression profile change in serums of patients with EOC during the treatment. Methods Sera of 11 patients with EOC were collected before operation, after operation, and after chemotherapy, respectively and hybridized with the same protein antibody chips. The results of the differentially expressed proteins were compared among these three groups. Results The number of detected differentially expressed proteins was decreased with increasing therapies. No same differentially expressed proteins was detected between these 3 groups. Eleven abnormally expressed proteins related to cell apoptosis were explored in these 3 groups and reversely correlated with the therapies. The synchronized abnormal expressions (up-regulated first and then down-regulated) of XPA and M cm2 might be related to drug resistance and recurrence. Conclusions Surgery and chemotherapy can result in the changes of protein expression in the serum of patients with EOC. The value of the changes of differentially expressed proteins in the serum during the treatment requires further investigation.
