摘要
目的:探讨同一温度不同时间和同一时间不同温度加热对凋亡坏死、survivin、caspase基因表达的影响与作用机制。方法:对数生长期胃癌AGS细胞,分别在同一温度不同时间和同一时间不同温度加热,用流式细胞仪检测细胞凋亡百分比和坏死百分比,用RT-PCR检测survivin和caspase基因的表达。结果:加热后细胞凋亡增加,加热温度42℃和加热时间2 h最为明显。随加热温度升高和加热时间增加坏死增加。加热明显加强Casepase-3的表达和降低Survivin的表达,且42℃加热不同时间时,2 h效果最明显,不同温度加热1 h时42℃效果最明显。结论:加热能明显抑制胃癌细胞生长,增强Casepase-3的表达和降低Survivin的表达,促进胃癌细胞的凋亡;加热温度42℃、加热时间2 h为胃癌细胞热疗的最佳条件。
Objective :To observe the effects of gastric cancer AGS cells on the apoptosis,cellular necrosis and expression of Survivin and casepase-3 gene by hyperthermia of different times at the same temperature and different temperatures at the same time and their mechanisms. Methods:Gastric cancer AGS cells at exponential phase of growth were heated at the same temperature for different times and for the same time at different temperatures. After dealing, cell apoptosis and cellular necrosis condition were detected by flow cytometry (FCM). To detecting apoptosis-related gene expression of casepase-3 and survivin by RT-PCR. Results:Cell apoptosis was increased after hyperthermia,which was most obvious after heating for 2h at 42 ℃. With the temperature and time increasing in hyperthermia, the cellular necrosis was increased.Hyperthermia could enhance the expression of casepase-3 and reduce the expression of survivin significantly. 2 h heating at 42 ℃ had the best result. Conclusion: Hyperthermia can inhibit the growth of gastric cancer, enhance the casepase-3 expression,reduce the survivin expression obviously and promote apoptosis of cancer cells. 2h heating at 42 ℃ is the best condition of hyperthermia.
