和厚朴酚联合青蒿素对CNE-2细胞增殖和凋亡的作用 被引量:5

Experimental of Honokiol and Artemisinin on Proliferation and Apoptosis of CNE-2 Cells

摘要 目的:研究和厚朴酚、青蒿素及二者联合应用在体外能否抑制人鼻咽癌CNE-2细胞的生长以及对其细胞凋亡的影响。方法:MTT法检测和厚朴酚、青蒿素单独或联合对细胞生长的抑制作用,以IC50评判抗肿瘤的效果,应用金氏公式进行联合用药的结果分析;DAPI染色观察细胞的凋亡形态,流式细胞仪检测和厚朴酚、青蒿素单独或联合作用对CNE-2凋亡率的影响。结果:和厚朴酚能显著抑制CNE-2细胞的增殖,并呈剂量依赖关系,其半数抑制浓度(IC50)为8.58 mg.L-1;青蒿素对CNE-2细胞生长没有明显的抑制作用,半数抑制浓度(IC50)为62.24 mg.L-1;和厚朴酚与青蒿素联合应用对CNE-2细胞的增殖抑制作用优于各单药组,q>1.15。DAPI染色可观察到和厚朴酚与青蒿素联合组核碎裂、凋亡小体,而单药组则不明显;凋亡分析显示联合用药组作用48 h细胞凋亡率显著高于各单药组。结论:和厚朴酚单独能够杀伤CNE-2细胞,并呈量-效关系,青蒿素单独作用不杀伤CNE-2细胞,两者联合应用对CNE-2细胞具有协同杀伤作用。 Objective: To investigate honokiol,artemisinin or combination of honokiol and artemisinin on proliferation and apoptosis of CNE-2 cells in vitro.Method: MTT assay was used to determine the effect of honokiol,artemisinin or their combination on the proliferation of CNE-2 cells.King formula was applied to analyze the combination effect of the two drugs.DAPI staining was used to observe the morphological changes of CNE-2 cells treated with honokiol,artemisinin or their combination for 48 hours.Flow cytometric analysis was used to detect the apoptosis rate of CNE-2 cells.Result: Honokiol could significantly inhibit the proliferation of CNE-2 cells,but artemisinin could inhibit the proliferation of CNE-2 cells slightly.The inhibitory effect of honokiol was in a dose-dependent manner.IC50 value of honokiol and artemisinin was 8.58,62.24 mg·L-1 respectively.The inhibition rate in combination group was much higher than that of the single drug group.Much more chromatin condensation,nuclear fragmentation and formation of apoptotic bodies was observed in combination group than single drug groups under fluorescence microscope.Compared with single drug groups,combination group induced higher apoptotic rate of CNE-2 cells by FCM analysis.Conclusion: Honokiol could inhibit proliferation of CNE-2 cells,while artemsinin had little effect on the proliferation of CNE-2 cells.However,honokiol combined with artemisinin yield a synergistc inhibitory effect on CNE-2 cells.
