系统性红斑狼疮患者血液中PRDM1和CD138+浆细胞表达的研究 被引量:2

摘要 目的;探讨系统性红斑狼疮(SlE)患者和正常人之间PRDM1和CD138+浆细胞的差异。方法:40例SLE患者血液标本,30例健康体检血液标本,抽取RNA、利用逆转录聚合酶链反应(RT-PCR)技术研究PRDMl基因表达;收集外周血单个核细胞,利用流式细胞技术研究CDl38’浆细胞量的差异。结果:SLE患者组血液中的PRDM1表达量明显高于健康体检组(p<0.05),并且CD138+浆细胞的量也多于健康体检组。结论:SLE实验组中的PRDM1明显高于体检对照组,CD138+浆细胞的量也高于对照组,提示SLE患者中浆细胞数量的增多与PRDMl高表达相关,这为进一步了解SLE发病机制提供了一个新的方向和靶点。 Objective: To determine the difference of PRDM - 1 and CD138 + plasma cells in between patients with systemic lupus erythematosus (SEE) and normal. Methods: forty cases of SLE patient blood samples, thirty cases of healthy blood samples, extract RNA, using reverse transcription polymerase chain reaction (RT- PCR) technique to study PRDMI gene expression; collection of peripheral blood mononuclear cells, differences in the amount of CD138+ plasma ceils by flow cytometry. Result: The PRDM1 expression in SLE patients blood was significantly higher than that healthy group (p 〈 0.05), and the amout of CD138+ plasma cells are also more than healthy group. Conclusion: PRDMI and the amount of CD138+ plasma cells in SLE study group is significantly higher than control group, indicating that the high PRDM1 expression and the number of plasma cells are relevant, this will provides a new direction and target to study SLE.
