茵栀黄注射液等28种中药制剂对大鼠体外CYP1A2的作用 被引量:4

摘要 目的:观察茵栀黄注射液等28种中药制剂对大鼠CYP1A2的作用。方法:以非那西丁为探针,通过研究其体外转化率的变化评价受试中药制剂对大鼠CYP1A2的影响。结果:清开灵注射液、茵栀黄注射液、穿心莲注射液使非那西丁的体外转化率分别为(255.87±32.33)pmol.min-1.mg-1、(204.01±43.60)pmol.min-1.mg-1、(220.15±48.39)pmol.min-1.mg-1,显著低于对照组(432.68±69.71)pmol.min-1.mg-1(P<0.05)。3种药物的IC50分别为3.55,2.43,2.49 mL.(100 mL)-1。结论:清开灵注射液、茵栀黄注射液、穿心莲注射液体外对大鼠CYP1A2有抑制作用,且呈浓度依赖性。 OBJECTIVE To evaluate the effects of 28 kinds of traditional Chinese medicine on rat liver CYP1A2 activity in uilro. METHODS The activity of CYP1A2 in rat liver microsomes was evaluated by detecting turnover of its substrate-phenac etin after treatment with the traditional Chinese medicine in vitro by HPLC. RESULTS The turnover of phenacetin treated by Qingkailing Injection, Yinzhihuang Injection and Chuanxinlian Injection was (255.87 ±32. 33)pmol.min^-1 mg^-1 , (204.01 ± 43.60)pmol.min ^-1.mg^-1 , (220. 15 ±48. 39)pmol.min^-1.mg^-1 respectively, which were significantly lower than that (432. 68 ± 69. 71 )pmol.min^-1.mg^-1 in the control group, (P〈0. 05). The IC50 values were 3.55, 2.43, 2.49 mL. 100(mL) ^-1' , re spectively. CONCLUSION Qingkailing Injection, Yinzhihuang Injection and Chuanxinlian Injection have significant inhibitory effects on the activity of rat CYP1A2 in a dose-dependent manner in vitro.
