MGMT在肾细胞癌中的研究

摘要 目的探讨O6-甲基鸟嘌呤-DNA甲基转移酶(O6-methylguanine-DNA-methyltransferase,MGMT)在肾透明细胞癌中的表达及意义。方法采用免疫组织化学法检测80例肾细胞癌组织及30例癌旁组织和20例正常肾脏组织中的MGMT表达,分析其与临床病理的相关性及意义。结果 MGMT在肾细胞癌组织中的阳性表达率为32.5%(26/80)明显低于癌旁组织76.7%(23/30)和肾脏正常组织65.0%(13/20),P=0.000;肾细胞癌组织高分化的阳性表达率47.1%(16/34)明显高于中分化28.6%(10/35)和低分化9.1%(1/11),P=0.034;肾细胞癌临床分期中T1+T2的表达(48.6%,18/37)明显高于T3+T4(18.6%,8/43),P=0.004。但与患者性别、年龄均无明显相关性。结论 MGMT可能成为肾细胞癌诊断及判断预后的标志物。 Objective To explore the expresstion and significance of O6-methylguanine-DNA-methyhrans- ferasegene (MGMT) in renal cell carcinoma (RCC). Methods The MGMT gene expression in 80 cases of RCC tissues and 30 case adjacent tissues and 20 cases normal renal tissues were detected by immunohistochemical method and the correlation between the expression and clinical factors were analyzed. Results The expression rates of MGMT in RCC 32.5% (26/80) was significantly lower than the normal renal tissue 65.0% ( 13/ 20) and adjacent tissues 76.7% (23/30) ,P =0. 000. The expression rates of MGMT in high grade 47.1% ( 16/ 34) was significantly hihger than middle grade 28.6% ( 10/35 ) and low grade 9.1% ( 1/11 ), P = 0. 034 ; The expression rates of MGMT in T1 + T2 48.6% ( 18/37 ) was significantly higher than in T3 + T4 18.6% (8/43) ,P = 0. 004, but the expressions of MGMT had no relationship with sex, age of RCC ( P 〉 0.05 ). Conclusions MGMT protein can be as a marker to diagnose and predict renal cell carcinomas.
