柳叶蜡梅提取物对人胃癌SGC-7901细胞的作用机制研究

Inhibitory effect of extract of Chimonanthus salicifolius on SGC-7901 cell and the possible mechanism

目的:探讨柳叶蜡梅提取物在体外对人胃癌细胞株SGC-7901的增殖抑制作用及其可能的机制。方法:应用细胞培养、噻唑蓝(MTT)比色法、光镜观察、扫描和透射电镜观察、Hoechst33258/PI染色等技术研究柳叶蜡梅提取物对SGC-7901细胞的抑制作用。结果:柳叶蜡梅提取物可明显抑制SGC-7901细胞生长,随着药物浓度的增加,呈现剂量依赖性效应;光镜及扫描透射电镜观察显示,药物处理组细胞表面微绒毛减少并发泡、细胞核固缩、染色质凝聚等形态学特征;细胞核经Hoechst33258染色出现颗粒状荧光,而PI染色无红色荧光出现。结论:柳叶蜡梅提取物对人胃癌SGC-7901细胞具有明显的抑制作用,其机制可能与细胞凋亡相关。

Objective： To investigate the inhibitory effect of extract of Chimonanthus salicifolius on human gastric cancer cell line SGC-7901 and the related mechanism.Methods： The effects of extract of Chimonanthus salicifolius on the cell proliferation of SGC-7901 cells were determined with MTT,light microscopy,electron microscopy,Hoechst 33258/PI staining.Results： The results showed that extract of Chimonanthus salicifolius treatment could significantly inhibit the growth of SGC-7901 cells in a dose-dependent manner.Light microscopy and electron microscopy showed that the morphology of the treated cells appeared less microvilli,blebbing,karyopyknosis,chromatin condensation.Cell nucleus concentrated and appeared granular fluorescence by Hoechst 33258 staining.Conclusion：This study suggested that Chimonanthus salicifolius has significant inhibitory effects on SGC-7901 cells proliferation,the possible mechanism might be related to cell apoptosis.
