红景天对急性心肌梗死大鼠心肌组织肾上腺髓质素及其受体表达的影响 被引量:13

Effects of Rhodiola on expression of adrenomedullin and calcitonin receptor-like receptor in myocardium of rats with acute myocardial infarction

摘要 目的:观察红景天对心肌梗死大鼠心肌组织肾上腺髓质素(AM)及其受体降钙素受体样受体(CRLR)表达的影响,探讨红景天促进大鼠缺血心肌血管新生的可能机制,了解红景天苷是否为红景天该作用的有效成分。方法:建立Wistar大鼠急性心肌梗死模型,造模前7d开始分组灌胃生理盐水、红景天或红景天苷,灌胃持续至造模手术后第7天,处死动物取心脏标本,进行HE染色,免疫组化测定梗死心肌边缘区和非梗死区CD31阳性反应强度,实时定量聚合酶链式反应法和蛋白印迹法检测心肌组织AM、CRLR的表达。结果:红景天组梗死边缘区心肌CD31表达显著高于生理盐水组(P<0.05);红景天组心肌组织AM、CRLR表达均明显高于生理盐水组(P<0.05),红景天苷组各指标的表达水平均介于生理盐水组和红景天组之间。结论:红景天具有上调AM、CRLR表达和促进AMI大鼠缺血心肌血管新生的作用,红景天苷是红景天发挥该作用的有效成分之一。 Objective: To investigate the effects of Radix et Rhizome Rhodiolae and Salidroside on expressions of adrenomedullin(AM) and calcitonin receptor-like receptor(CRLR) in myocardium of rat with acute myocardial infarction(AMI),to elucidate the possible mechanism of Radix et Rhizome Rhodiolae in promoting,and to investigate that whether or not salidroside could be considered as the effective component of Radix et Rhizome Rhodiolae with regard to the effects mentioned above.Methods: Four groups of male Wistar rats were studied: twenty-seven rats ligated as AMI model were fed with natural salt(N group),Rhodiola solution(R group) or Salidroside solution(S group) from 7 days before to 7 days after operation.Another three rats were randomized to normoxia control group(C group).All rats were sacrificed 7 days after the operation.Immunohistochemistry assay(IHC) was used for detecting the expression of CD31;SYBR Green real-time quantitative polymerase chain reaction(PCR) was employed for detection of the levels of AM and CRLR mRNAs;Western Blot analysis was used for detection of theirs protein levels.Results: Results of IHC index showed that the expression of CD31 at infarct border zone was significantly increased in R group as compared with the other groups,and S group took the second place(P〈0.05).The expressions of AM and CRLR mRNAs and proteins were significantly increased in R group as compared with N group(P〈0.05).All the expression levels,including those of AM and CRLR,in S group were higher than those in N group while lower than those in R group.Conclusion: Pretreatment of Rhodiola or Salidroside can promote angiogenesis in rat ischemia heart through elevating the expressions of AM and CRLR.Salidroside may be one of the effective components in Radix et Rhizome Rhodiolae.
