同型半胱氨酸转甲基酶的制备及纯化

摘要 目的建立同型半胱氨酸转甲基酶(BHMT)的制备及纯化方法,为临床应用提供基础。方法采用大肠埃希菌表达体系获得BHMT蛋白,然后应用Sephadex CL-6B、DEAE Sepharose Fast Flow、Sephadex G-75层析柱纯化BHMT,并行Western blot验证。分别用商品化同型半胱氨酸试剂盒和提纯的BHMT测定20份血清的同型半胱氨酸浓度,初步判断纯化的BHMT活性。结果 (1)BHMT在大肠埃希菌体系中成功表达,并经Sepha-dex CL-6B、DEAE Sepharose Fast Flow、Sephadex G-75层析柱纯化后在Western blot上显示为单一条带,分子量为45 kD。(2)采用纯化的BHMT检测20例标本,与商品化试剂盒检测结果差异无统计学意义[(17.19±4.10)μmol/L vs(16.48±3.48)μmol/L,P>0.05],且结果呈高度正相关(r=0.92,P<0.05)。结论建立的BHMT的制备及纯化方法切实可行,有望应用于同型半胱氨酸浓度的检测。
