ω-3多不饱和脂肪酸对人外周血树突状细胞的影响 被引量:1

摘要 目的探讨ω-3多不饱和脂肪酸对树突状细胞成熟状态及免疫学功能的影响。方法取新鲜健康成人外周血,经淋巴细胞分离液梯度分离获得单个核细胞,经含重组人粒细胞-巨噬细胞集落刺激因子(rhGM-CSF)和重组人细胞白介素-4(rhIL-4)诱导人外周血单核细胞(PBMC),获得未成熟DCs,将其分为5组:未成熟DCs组、成熟DCs组、二十碳五烯酸(EPA)、二十二碳六烯酸(DHA)及SFA处理组。经EPA、DHA孵育24 h,内毒素脂多糖(LPS)作用48 h。Wright-Giemsa染色观察各组DCs形态;采用半定量反转录-PCR(RT-PCR)检测DCs共刺激分子CD80、CD86及表面抗原提呈分子人类白细胞抗原-DR(HLA-DR)mRNA的表达;采用ELISA法检测各组细胞上清中白细胞介素-12(IL-12)及肿瘤坏死因子-α(TNF-α)的含量。结果形态学可见成熟DCs组诱导明显,未成熟DCs组、EPA、DHA组伪足少且细胞粗大;细胞上清IL-12和TNF-α的含量,EPA、DHA处理组比成熟DCs组低(P<0.01),而SFA组与成熟DCs组相比差异无统计意义(P>0.05);CD80、CD86及HLA-DR的mRNA相对表达量,成熟DCs组比未成熟DCs组表达量高(P<0.01),EPA、DHA处理组比成熟DCs组表达量低(P<0.01),而SFA处理组与成熟DCs组相比差异无统计学意义(P>0.05)。结论ω-3多不饱和脂肪酸可下调DCs共刺激分子CD80、CD86及HLA-DR的表达,降低其IL-12及TNF-α的释放,减弱DCs的抗原提呈能力,且对DCs抑制体外成熟。
