裂褶菌孢内多糖和孢外多糖对小鼠免疫功能的影响 被引量:26

摘要 裂褶菌孢内多糖(SPG_1)和孢外多糖(SPG_2)是分别从发酵培养的裂褶菌菌丝体和发酵液中提取的多糖。SPG_1和SPG_2均能显著促进Con A诱导的小鼠脾淋巴细胞增殖反应,SPG_1还显著地对抗氢可的松对淋巴细胞增殖反应的抑制作用。SPG_1和SPG_2均可恢复14月龄老年小鼠低下的脾淋巴细胞增殖反应。给小鼠ip SPG_1和SPG_2均能显著增强二硝基氯苯(DNCB)所致小鼠迟发型皮肤过敏反应(DCH)。小鼠ip SPG_1可显著增强羊红细胞(SRBC)诱导的小鼠脾脏空斑形成细胞(PFC)反应。SPG_1和SPG_2还可使老年小鼠的PFC反应恢复至成年小鼠水平。 SPG_1 and SPG_2 are polysaccharides extracted respectively from fermented cultural mycelium and fermentation fluid of Schizophyllum commune. This paper reports the effect of SPG_1 and SPG_2 on concanavalin A (Con A) induced proliferation of lymphocyte from C_(57)BL/6J mice in vitro. SPG_1 (10, 25, 50 and 100 μg/ml) and SPG_2 (10, 50 and 100μg/ml) was found to significantly increase lymphocyte proliferation. In addition, SPG_1 (10, 25, 50 and 75μg/ml) antagonized the suppressive effect of hydrocortisone on proliferation of mouse spleen cells, while SPG_2 did not. In vivo, at the dose of 2.5 and 5.0 mg/kg ip for 9 days, SPG_1 and SPG_2 remarkably enhanced the delayed-type hypersensitivity to 2, 4-dinitrochlorobenzene (DNCB) in mice. SPG_1 (1.25, 2.5 and 5.0mg/kg, ip for 1 day) and SPG_2 (2.5 and 5.0 mg/kg, ip for 1 day) promoted the plaque forming cell(PFC) response to sheep red blood cells (SRBC) in mice. The effects of SPG_1 and SPG_2 on immune function were studied in aged immunodeficient mice. SPG_1 (10 and 25 μg/ml) and SPG_2 (25 μg/ml) was found to enhance the Con A induced lymphocyte proliferation in 14 months aged mice. At the dose of 2.5 mg/kg, SPG_1 and SPG_2 produced respectively 162.1 and 136.0% increase in PFC response in 14 months aged mice.
