解脲脲原体生物群在生物膜形成能力及生物膜形成后耐药性的差异 被引量:8

Differences in the Ability of Biofim Formation and Antibiotics Resistance after Biofilm Fromation between Biovar of Ureaplasma Urealyticum

摘要 目的观察解脲脲原体(Uu)Parvo和T960两大生物群在体外形成生物膜的能力及生物膜形成后药物敏感性的差异。方法以Uu标准株3和8血清型及从21个女性患者宫颈分离出的21株Uu临床株为研究对象。以多条带抗原(MB-Ag)基因为靶位设计分群引物进行Uu生物分群检测。体外培养生物膜后扫描电镜及共聚焦激光显微镜鉴定生物膜形成,结晶紫半定量法检测生物膜形成能力,以微量肉汤稀释法对菌株生物膜形成前后进行药敏测定(四环素,红霉素和环丙沙星)。结果 Uu两生物群均具有生物膜形成能力,在生物膜形态及形成量上无明显差异,生物膜形成后对3种药物的耐药性均增高(P<0.05),但Pavor生物群变化更大。结论 Uu两生物群在生物膜形态及形成量上无明显差异,但形成生物膜后耐药性可能存在差异。 Objective To investigate the biofilm-forming ability and the difference of antibiotic susceptibility between biovar Parvo and T960 of Ureaplasma urealyticum after biofilm developed in vitro. Methods Total 23 strains of U. urealyticum have been tested, which included one U. urealyticum serovar 3, one serovar 8 reference strain and 21 clinical strains collected from female patients with. cervicitis. Ureaplasma biovars determination was performed by gene amplification of the multiple-banded antigen (MB-Ag). Scanning electron microscope and confocal laser scanning microscopy were used to analysis the biofilms formation. The biomass of each biofilm was semi-quantified by using a crystal violet staining technique. The broth microdilution susceptibility tests for tetracycline, erythromycin and ciprofloxacin were carried out before and after the biofilm formation. Results Both biovar Parvo and biovar q960 can form the biofilms in vitro with no significant difference in morphology and quantity. The sessile cells in biofilms were more resistant to the 3 antibiotics tested than the planktonic cells before the biofilm developed in these two Ureaplasm biovar groups (P 〈 0.05 ), in which the difference within biovar Parvo was more significant. Conclusion There is no significant difference in mophology and quantity of biofilms formation between the biovar Parvo and 3960 of Ureaplasma urealyticum, but both of them developed antibiotic resistance to certain antibiotic after biofilm formation.
