Hhcy及B族维生素干预对大鼠脑血管ICAM-1表达的影响

摘要 目的:探讨高同型半胱氨酸血症(Hhcy)及叶酸、维生素B12和维生素B6干预对大鼠脑皮层血管细胞间黏附分子-1(ICAM-1)表达的影响。方法:将40只雄性SD大鼠随机分为对照组、Hhcy组、治疗组和预防组,采用不同饮食结构及干预手段制备动物模型。采用高效液相色谱(HPLC)荧光检测技术测定不同时期各组大鼠血浆总同型半胱氨酸(tHcy)水平,并于第12周末通过免疫组化法观察ICAM-1在大鼠脑皮层血管的表达情况。结果:第4周末大鼠Hhcy模型制备成功,而第12周末对照组、治疗组和预防组血浆tHcy水平差异无统计学意义;预防组大鼠脑皮层ICAM-1阳性血管数与对照组差异无统计学意义,余各组比差异有统计学意义(P<0.05或P<0.01)。结论:Hhcy可以促进大鼠皮层脑血管ICAM-1表达,B族维生素可以有效降低血浆tHcy水平,并且对ICAM-1表达有一定的干预作用。 Objective: To observe the effects of hyperhomocysteinemia (Hhcy), folic acid, Vitamin B12 and Vitamin B6 on the expression of intercellular adhesion molecule 1 (ICAM-1) in the brain of rats with Hhcy. Methods:Forty healthy male Sprague-Dawley rats were divided into 4 groups, control group, Hhcy group, treatment group and prevention. The rat model was established by the way of different foods and different interventions. The plasma levels of total homocysteine (tHcy) were assayed by high-performance liquid chromatography method (HPLC) in different time points in groups. At the end of the 12th week, the expression of ICAM-1 in cerebral vessels of rats was detected by immunohistochemical method. Results: The Hh- cy model was induced successfully after 4 weeks. There were no statistical differences in plasma levels of tHcy between treat- ment group, prevent group and control group at the end of 12 weeks. There were no statistical differences in ICAM-l-positive cerebral vessels between rats of prevent group and control group, but there were significant differences between rats of other groups (P 〈 0.05 or P 〈 0.01). Conclusion: The expression of ICAM-1 was up-regulated by Hhcy in cerebral vessels of rats. The intervention of B Vitamins could decrease the plasma level of tHcy effectively and reduce the expression of ICAM-1 in cerebral vessels of rats.
