辅助伴侣蛋白STUB1及其功能研究进展

摘要 STUB 1分子是一类新型的辅助性伴侣蛋白,在蛋白质折叠、组装、转运和降解中起着重要的调节作用,属于泛素连接酶。STUB 1具有E3泛素连接酶活性,能与Hsp70、Hsp90或者其它分子伴侣结合,促进底物的连接和链的延伸。STUB 1具有调控阿尔采末病、帕金森病、麦-考二氏综合征等神经退行性疾病相关的tau蛋白、Aβ、α-突触核蛋白、MKKS突变体等降解的作用;同时STUB1还受辅助伴侣蛋白HspBP1及细胞激酶Akt的调控。现将辅助伴侣蛋白STUB 1及其在不同疾病中的调节功能研究进展综述如下。 STUB1 is a new molecular co-chaperones,and plays an important role in regulating protein folding,assembly,transport,and degradation.STUB1 belongs to the ubiquitin ligase.STUB1 shows E3 ubiquitin ligase activity,and can combinte with Hsp70,Hsp90 or other molecular chaperones.It promotes the connection between substrates and chain extension.STUB1 regulates the protein degradation of neurodegenerative diseases,such as tau protein and Aβ of Alzheimer's disease,α-synuclein of Parkinson's disease,MKKS mutants of McKusick-Kaufman syndrome,while STUB1 is regulated by both chaperone protein HspBP1 and cell kinase Akt.
