TLR2在HSE小鼠脑组织中的表达及其免疫损伤 被引量:1

Preliminary study on the expression and effect of TLR2 in the mouse tissues of herpes simplex encephalitis

摘要 检测TLR2在单纯疱疹病毒脑炎(HSE)小鼠脑组织中表达变化,为探讨HSE的免疫学发病机制提供实验资料。采用颅内注射单纯疱疹病毒1型(HSV-1)建立HSE小鼠模型,并设置盐水组和正常对照组。鉴定造模成功后,应用RT-qPCR方法检测各组小鼠脑组织TLR2 mRNA的相对表达;常规制备病理石蜡切片,HE染色后观察HSE小鼠脑组织病理改变。结果HSE小鼠脑组织TLR2 mRNA的相对表达明显高于盐水组和正常组(P<0.01),HSE小鼠脑组织出现充血、坏死等病理改变。表明TLR2可能在HSE发病的免疫学机制中起着重要作用。 To investigate the TLR2 expression in the brain of mice infected with herpes simplex encephalitis(HSE) in order to provide the experimental data for the investigation of the immunological pathogenesis of HSE,the mouse model of HSE was established through introcalvarium inoculation of herpes simplrx virus-1 type(HSV-1),meanwhile saline group and normal group were set up.After this mouse model were built successfully,TLR2 expression in mouse brain of each groups were detected by RT-qPCR and the pathological changes were observed in brain tissue of mice HSE through routine paraffin section preparation and HE staining.It was found that TLR2 expression in HSE group were increased obviously than those of other groups(P0.01),and congestion,necrosis and other pathological changes could be demonstrated in mouse brain tissues infected with HSV-1.It is evident that TLR4 may play an insignificant role in immunology pathogenesis of HSE.
