丙戊酸钠对体外培养许旺细胞增殖及相关蛋白GAP-43、MAG和MBP表达的影响 被引量:4

摘要 目的观察丙戊酸钠(VPA)对体外培养的大鼠坐骨神经许旺细胞的影响,探讨其促进周围神经再生的作用机制。方法取sD幼鼠坐骨神经体外培养许旺细胞,应用免疫荧光技术鉴定许旺细胞纯度。将体外培养的许旺细胞分为实验组和对照组,分别给以含与不含VPA(1.0mmol)的血清培养液,处理后分别于0、12、24、36、48和72h采用4-甲偶氮唑蓝(MTT)法检测细胞增殖,免疫组化法检测雪旺细胞特异性蛋白(S-100)、生长相关蛋白(GAP-43)、髓鞘相关蛋白(MAG)和髓鞘碱性蛋白(MBP)表达,以及蛋白免疫印迹(Western blotting)法检测GAP-43。MAG和MBP表达。结果与对照组相比,在24、36h和48h实验组许旺细胞增殖率显著增加,以48h两组差别最为明显(P〈0.01);在24、36h和48h实验组GAP-43和MAG蛋白表达水平显著增加(P〈0.05);在36h和48h实验组MBP蛋白表达水平显著增加(P〈0.05)。结论临床使用剂量的VPA促周围神经再生的作用机制可能与其诱导许旺细胞增殖、上调GAP-43、MAG和MBP蛋白表达水平有关。
