卵巢癌组织AM表达及其对ERK活性影响的观察 被引量:3

Expression of adrenomedullin in ovarian cancer and its influence to ERK activity

摘要 目的:研究卵巢上皮性恶性肿瘤中肾上腺髓质素(AM)的表达及其对ERK活性的影响,探讨其作用机制及临床意义。方法:应用RT-PCR、MTT比色法和蛋白质印迹法对卵巢上皮性恶性肿瘤(30例)、卵巢良性肿瘤(10例)和正常卵巢组织(10例)的AM表达情况及AM、AM受体阻断剂(AM22-52)对卵巢癌细胞增殖和ERK活性的影响进行分析。结果:AM在卵巢上皮性恶性肿瘤中的半定量结果为0.948±0.150,与正常卵巢及卵巢良性肿瘤比较,差异有统计学意义,P=0.042。其表达量与术后病理分级相关,与手术病理分期无关;MTT法检测显示,AM能促进卵巢癌细胞增殖的作用,随着作用浓度和作用时间的增加这种促进作用也随之增强;蛋白质印迹法检测,随着AM作用浓度的增加,磷酸化的ERK1/2的表达随之增高,反之,随着AM受体阻断剂(AM22-52)浓度的增加,磷酸化的ERK1/2的表达随之降低,差异有统计学意义,P<0.05。结论:卵巢癌组织中AM高表达,其能通过MAPK/ERK信号转导途径在卵巢癌的发生中起作用。 OBJECTIVE: To evaluate the expression level of AM in epithelial ovarian cancer and its influence to ERK activity,and detect the interaction mechanisms and clinical significance.METHODS: By using RT-PCR,MTT and Western blot the expression level of AM in 10 cases normal ovarian tissues,10cases benign ovarian epithelial tumors,and 30 cases malignant ovarian epithelial tumors were analyzed,and the influence of different density AM and the AM acceptor blocking agent(AM22-52) on the ovarian cancer cell proliferation and ERK activity change were observed.RESULTS:The semi-quantitative results showed that the level of AM was much higher in the malignant ovarian epithelial tumors(0.948±0.150) than that in the normal ovarian tissue or benign ovarian tumor(P=0.042).There was significantly difference in the different histological grade,but not in the different clinical stage.The result of MTT showed that AM could promote the ovarian cancer cell multiplies,which was direct correlated with the AM concentration and reactive time.Western blot method demonstrated AM could tone up the ERKI/2 activeness along with the AM density,while,AM acceptor blocking agent(AM22-55) could reduce it.CONCLUSION: AM is highly expressed in the ovarian cancers which may play a role in the development of ovarian cancer through the MAPK/ERK signal transduction pathway regulation.
