溶氧对灵芝液体深层培养产胞外多糖的影响 被引量:5

Effects of Dissolved Oxygen on Exocellular Polysaccharides Produced by Deep Liquid Cultivation of Ganoderma lucidum

摘要 以Canoderma lucidum xy05为生产菌,采用亚硫酸盐法测定了摇床上摇瓶不同装液量下摇床转速氧容积传递系数,比较了不同氧容积传递系数对菌丝生长和产胞外多糖的影响,确定氧容积传递系数控制在38~68 h-1时,发酵效果最佳。在发酵罐上比较了恒定发酵罐搅拌转速、恒定发酵液DO值和分段控制发酵液DO值3种不同控氧方式,结果表明,分段控制发酵液DO值效果最佳。在最佳的分段控氧模式下菌丝干重达10.88 g/L,胞外多糖达2.88 g/L。 Canoderma lucidum xy05 was selected to produce polysaccharides.The values of kLa of flask with different volume of liquid medium in different rotate speed of shaker were determined by method of sulphite.The effects of kLa value on the growth and fermentation of C.lucidum were compared,and the result showed the optimal kLa value was 38~68 h-1.The three methods of dissolved oxygen control in fermentor,fixed rotate speed of fermentor,fixed DO value of fermentor and subsection control of DO value of fermentor,were compared,and the result showed the subsection control oxygen mode was the optimal.In the best condition,the dry weight of hypha was 10.88g/L,the exocellular polysaccharides reached 2.88 g/L.
