鸡胚地龙膏对兔急性软组织损伤血清及组织液中疼痛指标的影响 被引量:12

摘要 目的观察鸡胚地龙膏对兔急性软组织损伤血清及组织液中疼痛指标的影响。方法以105只新西兰白兔为研究对象,随机选出21只作为正常对照组,剩余84只新西兰白兔造模成功后随机分为4组,分别为模型对照组、阳性药物组、鸡胚地龙膏低剂量组和鸡胚地龙膏高剂量组,每组21只动物。模型组不作任何处理,自然恢复愈合,阳性药物组采用奇正消痛膏治疗(0.75g/kg),鸡胚地龙膏低剂量组和鸡胚地龙膏高剂量组采用鸡胚地龙膏治疗,药物剂量分别为0.15g/kg和0.75g/kg。阳性药物组、鸡胚地龙膏低剂量组和鸡胚地龙膏高剂量组药膏涂布面积以超过受损区面积1/2以上为度,外用特制胶布及卫生胶布固定、包扎,每天换药1次,共给药8天。各组分别于损伤前,损伤后即刻,损伤后第1、2、3、6、8天抽取5ml静脉血后,处死动物,留取损伤部位组织,制作组织液。采用酶联免疫法分别测定不同时间血液和组织液中前列腺素E2(PGE2)、5-羟色胺(5-HT)和组织胺(HIS)的含量水平,并进行统计学分析。结果鸡胚地龙膏高剂量组从损伤后第2天开始血清及组织液中PGE2、5-HT、HIS逐渐降低,其血清及组织液中PGE2、5-HT、HIS含量水平低于鸡胚地龙膏低剂量组和阳性药物组(P<0.01)。伤后3天鸡胚地龙膏高剂量组血清及组织液中PGE2、5-HT恢复到损伤前水平,较鸡胚地龙膏低剂量组和阳性药物组提前5天恢复到损伤前的正常水平。而HIS含量伤后6天恢复正常,较鸡胚地龙膏低剂量组和阳性药物组提前2天。结论鸡胚地龙膏可有效降低兔急性软组织损伤后的血清和组织液中PGE2、5-HT和HIS等疼痛因子的含量水平,减轻损伤后疼痛程度。 Objective To observe the effects of Jipei ointment on pain indexes in serum and tissue fluid of rabbits with acute soft tissue damage.Methods 105 New Zealand white rabbits were used as the research objects.21 were randomized into the normal control group.The remaining 84 New Zealand white rabbits after the success of modeling were divided into four groups immediately: the model control group,the positive medicine group,the Jipei ointment low dose group,and the Jipei ointment high dose group(21 animals in each group).No treatment was given to the model group,which would recover naturally.Cheezheng pain relieving plaster(0.75 g/kg) was used in the positive medicine group.Jipei ointment was used in the Jipei ointment low dose group and the Jipei ointment high dose group(0.15 g/kg and0.75 g/kg,respectively).The ointment covering area of the positive group,the Jipei ointment low dose group and the Jipei ointment high dose group was more than 1/2 of their damaged area.Special and healthy strapping tapes were used to fix and wrap the wound.A dressing change was performed every day,with 8 days for administration.5 ml venous blood was collected from the rabbits in each group respectively before injury immediately,after injury immediately and 1,2,3,6,8 days after injury.Then the rabbits were killed,with the tissues of the damaged area left to make tissue fluid.The levels of PGE2,5-HT and HIS in respective blood and tissue fluid were measured by means of enzyme linked immunosorbent assay.Then statistical analysis was performed.Results On the first day after injury,the levels of PGE2,5-HT and HIS in blood and tissue fluid of the rabbits of the Jipei ointment high dose group gradually reduced,which was lower than those of the Jipei ointment low dose group and the positive medicine group(P0.01).Three days after injury,the levels of PGE2,5-HT and HIS in blood and tissue fluid of the rabbits of the Jipei ointment high dose group recovered to the level before the injury,which was 5 days earlier than those of the Jipei ointment low dose group and the positive medicine group.Six days after injury,the level of HIS returned to normal,which was 2 days earlier than the Jipei ointment low dose group and the positive medicine group.Conclusion Jipei ointment can effectively reduce the levels of PGE2,5-HT,HIS and other pain indexes in blood and tissue fluid of rabbits with acute soft tissue injury.It can relieve pain of injury.
