摘要
目的探讨糖尿病大鼠Ⅳ型胶原(Col Ⅳ)、纤维连接蛋白(FN)及核转录因子kappa B(NF-κB)蛋白的表达及苯那普利对其影响。方法将34只Wistar大鼠随机分为3组:正常对照组(C组)、糖尿病未治疗组(D组)、糖尿病加苯那普利治疗组(B组)。腹腔注射链脲佐菌素(STZ)诱导糖尿病模型。检查各组大鼠在灌胃给药第12周血糖、血肌酐、尿素氮、尿白蛋白及体重等指标变化。应用免疫组织化学检测Col Ⅳ、FN及NF-κB蛋白的表达水平。结果第12周时B组大鼠血糖、血肌酐的浓度及尿蛋白排泄率较D组明显减少(P<0.01<0.05);免疫组织化学显示,B组大鼠肾组织Col Ⅳ、FN及NF-κB蛋白的表达明显低于D组(P<0.01)。结论苯那普利对糖尿病大鼠肾脏具有部分保护作用,其可能抑制血糖浓度升高,下调糖尿病大鼠Col Ⅳ、FN及NF-κB蛋白的表达,减少细胞外基质聚积。
【Objective】 To investigate the mechanisms of Benazepril on the expression of type IV collagen(col Ⅳ),fibronectin(FN) and nuclear factor kappa B(NF-κB) in renal tissue of streptozotocin(STZ) induced diabetic rats.【Methods】 34 Wister rats were randomly divided into three groups: normal control group(C group,n =10),diabetes untreated group(D group,n =12) and diabetes treated with Benazepril(B group,n =12).Diabetes mellitus models were induced by injection of STZ intraperitoneally.Plasma glucose,serum creatinine,blood uria nitrogen,urinary albumin were observed respectively at the twelfth week of treatment,and the expression of type Ⅳ collagen(col Ⅳ),fibronectin(FN) and NF-κB in the kidney were measured by immunohistochemistry.【Results】 At the twelfth week,plasma glucose,creatinine and urinary albumin excretion in B group were significantly lower than those in D group(P 0.01).immunohistochemistry results showed that the expressions of type Ⅳ collagen,fibronectin and NF-κB in renal tissue in B group were much lower than those in D group(P 0.01).【Conclusions】 Benazepril has some renal protective effects on diabetic rats.This might result from decreasing plasma glucose level,down-regulating type Ⅳ collagen,fibronectin and NF-κB expression and reducing the deposition of glomerular extracellular matrix.
