摘要 目的观察辛伐他汀后处理对牛肺动脉内皮细胞缺血再灌注损伤的影响。方法对牛肺动脉内皮细胞行5 h的氧糖剥夺和1 h的再灌注。在再灌注之初立即将不同剂量的辛伐他汀加入到内皮细胞中,抑制剂组加入相应的抑制剂。观察不同剂量辛伐他汀组、氧糖剥夺组、抑制剂组及对照组中LDH的释放水平。结果辛伐他汀剂量依赖性地降低了氧糖剥夺和再灌注导致的乳酸脱氢酶释放,加入抑制剂可以逆转或消除他汀后处理的保护作用。结论辛伐他汀后处理可以减少氧糖剥夺和再灌注导致的内皮细胞损伤。 【Objective】 To observe the effects of Simvastatin postconditioning on ischemia reperfusion induced bovine pulmonary arterial endothelial cell(BPAEC) injury.【Methods】 BPAEC were subjected to a 5 h oxygen-glucose deprivation(OGD) and a 1 h simulated reperfusion.Simvastatin alone or plus mevalonate,geranylgeranyl pyrophosphate(GGPP) were added to cells immediately at the beginning of the simulated reperfusion.We observe lactate dehydrogenase(LDH) release of different groups.【Results】 Simvastatin dose-dependently reduced the OGD and simulated reperfusion-induced LDH release from primary BPAEC.This effect was inhibited by mevalonate,GGPP.【Conclusions】 These results suggest that statin post-treatment reduces OGD and simulated reperfusion-induced cell injury.
