摘要
目的:研究黄芩苷抑制静脉桥术后内膜增生的作用及其机理。方法:通过建立动脉粥样硬化兔模型,在此基础上建立颈外静脉-颈总动脉移植模型,实验过程中用黄芩苷(中药组)进行干预,同期用西药非诺贝特(西药组)进行对照干预。测定血清肿瘤坏死因子(TNF-α)及白细胞介素6(IL-6)含量,并检测静脉桥内膜、中膜厚度。结果:中药中、高剂量预防组,中药中、高剂量治疗组与西药预防、治疗组比较,能有效降低TNF-α、IL-6的含量(P<0.05);中药预防各组,中药中、高剂量治疗组与西药预防、治疗组比较,静脉桥内膜增生厚度明显降低(P<0.05);且呈剂量依赖关系,黄芩苷给药浓度越高,上述作用越明显。结论:黄芩苷具有减缓静脉桥血管内皮细胞和平滑肌的细胞增殖,从而起到抑制静脉桥内膜增生的作用。
Objective: To observe the effect of baicalin on inhibiting intimal hyperplasia in rabbits after vein graft, and to explore its mechanism. Methods: We reconstructed the common carotid artery with autogenous external jugular vein on atherosclerosis (AS) rabbit models. During the experiment, intervention of baicalin (treatment group) and fenofibrate (control group) was carried out. The contents of serum tumor necrosis factor-a (TNF-a) and interleukin 6 (IL-6) were assessed, and the thickness of grafted vein intima and media was measured. Results: Compared with the control group, the contents of serum TNF-a and IL-6 were decreased(P 〈 0.05), and the intimal thickness was reduced in the treatment group(P 〈 0.05), and the effect was in concentration-dependant manner. Conclusion: Baicalin can slow down the proliferation of endothelial cells and smooth muscle cells of grafted veins, which results in the inhibition of the intimal hyperplasia.
