摘要
目的探讨黑米花青素(black rice anthocyanins,BRACs)对大鼠肠道菌群及抗氧化能力的调节作用。方法 24只Sprague-Dawley大鼠正常饲养1周后,随机分成对照组(n=6)和BRACs组(n=18),其中BRACs干预剂量分别为50、100、150mg/kg.bw,每天灌胃1次,连续15d,对照组则灌胃等体积生理盐水。实验前后,采集各组大鼠新鲜粪便,平板计数法测定肠道双歧杆菌、乳杆菌、肠球菌和肠杆菌数量;最后取大鼠眼眶血测量其脂质过氧化产物丙二醛(malondialdehyde,MDA)含量和抗氧化酶系超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)和谷胱甘肽转移酶(glutathione S-transferase,GST)的活性。结果 BRACs连续灌胃15d后,BRACs组肠球菌和肠杆菌数量均比对照组明显降低(P<0.05),而双歧杆菌和乳杆菌数量与对照组相比则无显著差异(P>0.05),3个剂量组组间在4个菌群数量上均无显著差异(P>0.05);BRACs组MDA含量显著低于对照组(P<0.01),而50mg BRACs/kg.bw组MDA含量高于其余2个剂量组(P<0.05);BRACs组SOD、GSH-PX和GST活性均明显高于对照组,但不同剂量间无显著差异(P>0.05)。结论 BRACs灌胃对肠道菌群有良性调节作用,这种调节作用可能与其抗氧化能力有关。
Objective To investigate the effects of black rice anthocyanins(BRACs) on intestinal flora and antioxidant capacity of rats.Methods 24 Sprague-Dawley rats fed with basic diet for 1 week were randomly divided into control group(n=6) and BRACs groups(n=18).The BRACs groups were administered with BRACs by stomach intubation at a dosage of 50,100,150 mg/kg·bw for 15 days,repectively.Meanwhile,the control group was administered with equal volume physiological saline without BRACs.The numbers of Bifidobacterium,Lactobacillus,Enterobacillus and Enterococcus in intestine were surveyed respectively using fresh faeces before and after experiment.And the concentration of malondialdehyde(MDA),the activity of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px) and glutathione S-transferase(GST) in serum were measured after experiment.Results The numbers of Enterococcus and Enterobacillus were markedly decreased in BRACs groups(P0.05),and there were no obvious difference in the number of Bifidobacterium and Lactobacillus between BRACs group and control group(P0.05).The serum MDA level in BRACs groups were decreased markedly compared to control group(P0.01),and it was higher in 50 mg BRACs/kg.bw group than that in the other BRACs groups(P0.05).Furthermore,BRACs treatment significantly increased the activity of antioxidases SOD,GSH-Px and GST in serum(P0.05).Conclusion BRACs had beneficial effects on intestinal flora in rats,which may be due to its potent antioxidant capacity.
