摘要 [目的]研究孔雀石绿对蚕豆根尖细胞有丝分裂的影响。[方法]采用不同浓度的孔雀石绿溶液处理蚕豆根尖细胞12、24、36、48和72h,通过常规染色体压片技术,观察蚕豆根尖细胞有丝分裂,并计算各处理组的有丝分裂指数。[结果]随着处理时间的延长,有丝分裂指数呈M型变化趋势,说明蚕豆根尖对孔雀石绿有一个适应过程。随着孔雀石绿浓度的升高,有丝分裂指数逐渐升高,在浓度为0.50~1.00mg/L时,有丝分裂指数整体达到顶点;随着孔雀石绿浓度的进一步升高,有丝分裂指数有所下降,说明孔雀石绿在一定浓度范围内对植物生长有利。[结论]该研究可以为孔雀石绿的合理利用提供依据,并为评价孔雀石绿对环境的危害提供参考。 [Objective] The research aimed to study the effect of malachite green(MG) on mitosis of Vicia faba root tip cell.[Method] Different concentrations of malachite green were used to treat Vicia faba root tip cell for 12,24,36,48 and 72 h.The mitosis of Vicia faba root tip cell was observed by conventional chromosome compression technology,and the mitotic index(MI) of each treatment group was counted.[Result] As the treated time prolonged,MI presented the M-shaped variation trend,which illustrated that Vicia faba root tip had a adaptive process to MG.As the concentration of MG increased,MI gradually rose.When the concentration of MG was during 0.50-1.00 mg/L,MI was the maximum.When the concentration of MG further increased,MI decreased.It showed that MG was favorable for the plant growth in the certain concentration range.[Conclusion] The research could provide the basis for the reasonable utilization of MG and the reference for evaluating the environmental harm of MG.
