摘要
目的采用基因芯片技术筛选稳定转染HBx基因的HepG2肝癌细胞对人脐静脉血管内皮细胞的差异表达基因。方法①以人脐静脉血管内皮细胞(HUVEC)为细胞模型,通过transwell共培养技术,分别将肝癌细胞HepG2及稳定转染HBx基因肝癌细胞HepG2(HepG2-X)与HUVEC共培养。②分别提取细胞总RNA,通过基因芯片技术筛选差异表达基因。结果有643条基因差异表达明显,其中UBR3、ATXN3等19条基因经HBx干预后表达量明显上升,FOLR1、HSPs、ZNF等624条明显下降。结论利用基因芯片技术可以筛选HBx参与的肝癌血管生成差异表达基因,为肝癌的诊治、预防提供一定的理论依据。
Objective To screen the differentially expressed genes in human umbilical vein endothelial cells(HUVEC) related to hepatocellular carcinoma cell HepG2 after stable transfection with HBx gene using cDNA microarray. Methods ①Human umbilical vein endothelial cells were used for cell model.HepG2 and HepG2-X(transfection with HBx) were co-cultured with HUVEC by transwell.②Total RNA was extracted from HepG2 and HepG2-X cells,respectively.Differentially expressed genes were obtained using cDNA microarray. Results A total of 643 differentially expressed genes were obtained,among which,19 were up-regulated such as UBR3,ATXN3,and the other 624 were down-regulated such as FOLR1,HSPs and ZNF. Conclusion The results suggest that cDNA microarray technology may be effective to screen the related genes in hepatocellular carcinoma and provide a theoretical basis for diagnosing and treating hepatocellular carcinoma.
