摘要
目的了解八味獐芽菜散对CCl4所致小鼠急性肝损伤的保护作用,探讨其保护机制。方法 40只昆明种健康小鼠随机分为四组。八味獐芽菜散组予八味獐芽菜散(450mg/kg.d)灌胃,连续7 d;联苯双酯组予联苯双酯(11.3 mg/kg.d)灌胃,连续7 d;空白对照组与模型组予等量蒸馏水灌胃,连续7 d。除空白对照组外,其他三组于第7 d用10%CCl4花生油溶液(10mL/kg)于腹腔注射,24 h后采集血样,测定小鼠血清ALT、TNF-α、IL-10。结果八味獐芽菜散组与模型组比较,ALT、TNF-α水平明显降低(P<0.01),而IL-10水平升高(P<0.01),但与联苯双酯组比较无显著性差异。结论八味獐芽菜散具有抗CCl4肝损伤的作用,其机理可能与提高IL-10、抑制TNF-α水平有关。
Objective To investigate the protective effects and mechanism of eight kinds of swertia powder on the acute liver damage caused by carbon tetrachloride (CCl4 ) in mice. Methods 40 healthy kun Mint mice were randomly divided into four groups. The experimental group was feed with eight kinds of swertia powder, 450 mg/kg/d, for 7 days. The second group was feed with Biphenyl Double Ester, 11.3mg/kg/d for 7days. The blank control group and model group were feed with same amounts of distilled water. Three groups except blank control were intraperitoneal injected with CCl4 peanut oil solution (10mg/kg) at 7th day. Then the blood samples were taken after 24 h and the alamine aminotransferase (ALT), tumor necrosis factor-α( TNF-α) , intedeukin -10 (IL- 10) were detected. Results Compared with model group, the levels of ALT, TNF-α and IL- 10 were significantly decreased in eight kinds of swertia powder group(P 〈0.01 ), but the level of IL - 10 was increased (P 〈 0.01 ). There was no difference compared with Biphenyl Double Ester group. Conclusion Eight kinds of swertia powder has the effect of protecting liver damage caused by CCI4 and the mechanism may related to increasing the level of IL - 10 and inhibiting the level of TNF -α.
