摘要
目的建立一种大鼠脊髓慢性压迫性损伤模型。方法将慢性膨胀物于显微镜下植入大鼠T8∕9水平,随着压迫物的膨胀,逐渐形成不同程度的慢性压迫脊髓损伤实验动物模型。手术后通过BBB评分进行行为学功能评定,检测压迫段脊髓病理标本及通过电脑图像分析系统半定量检测凋亡启动基因P53。结果术后观察行为学、组织学、病理学上的改变,3者相符合。结论此模型较以往模型相比,具有制作简单,损伤程度可调整,脊髓损伤能表现出不同的压迫程度,可重复性强等优点。为进一步研究脊髓慢性压迫损伤病理机制奠定基础。
Objective The purpose of the present study was to establish an animal mode of chronic compressed spinal ,cord injury in the rat to explore possible mechanisms ofspinl cord chronic compress. Methods A water-swellable robber was implanted microsurgical underneath Ts/9 laminae of rat.The spinal cord were progressively compressed by the expan- ding of the waterswellable robber.neurological .Function evaluated by BBB,the compressed spinal cord was harvested and examined histopathologically at 1,3,7, 14 and 28days post-operation. The expression of apoptosis factors, P53mRNA was examined by hybridization in situ ,semiquantitave analysis by a computer image analysis system. Results The BBB scale,the P53mRNA positive expression and histopathological were coincidence of the character of chronic spinal cord compression myelopathy. Conclusion Techniques for creating this model are simple and can be highly replicable,and injury degress of spinal cord can indicate the compress depth .It provides an available basis for the further studies on pathological mechanism of chronic spinal compressed injury.
