摘要
从山东近岸海域海水中分离得到一株产黑色素细菌S1,通过对S1菌株的菌落形态观察、生理生化性质检验及16SrDNA序列测定,初步鉴定为明亮发光杆菌(Photobacterium phosphoreum)。为了提高S1菌株黑色素产量,采用单因素实验设计对S1菌株进行发酵条件优化,结果表明当发酵条件为L-酪氨酸0.75g/L、pH 7.5、盐度30、以占摇瓶1/5的培养量37℃下发酵培养时,S1菌株产黑色素的量最高。
A strain of melanin bacterium was isolated from Shandong costal seawater and was identified as Photobacterium sp.based on morphological,physiological and biochemical tests as well as the bacterial 16S rDNA sequence analysis.In addition,the condition for melanin formation was optimized by single factor experimental design in order to increase the bacterial melanin production.The results showed that the optimum condition was as follows:L-tyrosine 0.75%,pH 7.5,salinity 30,temperature 37℃ and the ratio of culture broth to vessel volume 1/5.
