5-羟甲基糠醛对过氧化氢致大鼠海马神经细胞损伤的保护作用研究 被引量:6

5-Hydroxyl Methyl-Furfural's Effect and Mechanism on Rat with H_2O_2 Induced Hippocampal Neurons Injuries

摘要 目的观察山茱萸活性成分5-羟甲基糠醛(5-HMF)对H2O2诱导损伤的大鼠海马神经细胞增殖、凋亡及LDH、SOD表达的影响。方法常规方法进行乳大鼠海马神经元的原代培养,建立体外H2O2氧化损伤模型,采用光镜和电镜进行形态学观察,MTT法对比不同浓度5-HMF对大鼠海马神经元增殖的影响,黄嘌呤氧化酶法检测各组细胞上清液中LDH、SOD的活力。结果光镜结果显示5-HMF大、中剂量组的细胞形态基本正常;电镜结果显示5-HMF大剂量组对细胞有一定程度保护作用。MTT结果显示5-HMF大、中剂量组OD值比H2O2模型组明显增高(P<0.05);LDH和SOD检测结果表明5-HMF大、中剂量组能明显降低培养液中LDH活性、升高SOD活性。结论山茱萸活性成分5-HMF大、中剂量组对H2O2损伤的大鼠海马神经元有一定保护作用。 OBJECTIVE To investigate the effect and mechanism of 5-hydroxylmethylfurfural(5-HMF),an active component extracted from Cornus officinalis on rat with hydroperoxide(H2O2)induced hippocampal neurons injures.METHODS Hippocampal neurons of newly born rat were cultured in vivo and were injured by H2O2.LM and EM were used to observe the morphological changes.Different concentration of 5-HMF on cell viability was measured by MTT;the activity of SOD was measured by xanthine oxidase.RESULTS It revealed that 5-HMF improved the morphology of H2O2-treated rat hippocampal neurons by LM and EM.The high and medium dosages of 5-HMF could decrease the activity of LDH,and increase the activity of SOD.CONCLUSION 5-HMF could protect the rat with H2O2 induced hippocampal neurons injures.
