高分辨率熔解曲线及其在分子诊断中的应用

摘要 通过加热使双链DNA解离为单链是DNA基本特性之一,高分辨率熔解曲线(high resolutionmelting,HRM)正是基于这一特性,采用荧光饱和染料监测熔链过程,从而获得DNA上的相关信息,例如突变、单核苷酸多态性(single nucleotidepolymorphism,SNP)等。由于其快速、高通量、低成本,已越来越多应用于临床疾病的分子诊断与分型。
