摘要 对多粘类芽孢杆菌20185发酵产碱性果胶酶的培养基和发酵培养条件进行了优化。经单因素和正交试验得到最优发酵培养基组成为(g/L):果胶10.0,蛋白胨20.0,K2HPO4.3H2O 6.0,KH2PO4 3.0,NaCl 10.0,MgSO4.7H2O 0.2,SDS 1.0。发酵最佳培养条件为:250mL三角瓶装液量为50mL,种龄24h,接种量9%,初始pH值9.0,温度35℃,培养36h。在此优化条件下,酶活为311U/mL。 The medium components and fermentation conditions for the alkaline pectinase produced by Paenibacillus polymyxa 20185 fermentation were studied. The optimized fermentation medium determined by single factors and orthogonal analysis methods was (g/L): pectin 10.0, tryptone 20.0, K2HPO4O3H20 6.0, KHEPO4 3.0, NaC1 10.0, MgSO4·7H2O 0.2, SDS 1.0. The corresponding optimum fermentation conditions were: 50ml liquid medium in a flask of 250ml, seed age 24h, inoculum 9%, initial pH value 9.0, culture temperature 35℃ and culture time 36h. Pectinase activity reached 311U/ml under the optimized medium and fermentation conditions.
