摘要
根据蒽醌类化合物在乙醇溶液中有较强的荧光性质,建立一种测定虎杖中总蒽醌的新方法。用80%vol乙醇溶液浸提,50℃超声提取虎杖中蒽醌类化合物,以1,8-二羟基蒽醌为标样,在激发波长lex=440nm,发射波长lem=520nm下,测定虎杖中总蒽醌的含量,并进行加标回收实验,验证方法的准确性。方法的检出限为0.15μg/mL,线性范围在0.3μg/mL^16.0μg/mL之间,线性回归方程y=179.27x+24.498,相关系数r=0.9993,平均回收率99.8%,相对标准偏差(RSD)0.87%。
A new method for determining total anthraquinone from Polygonum cuspidatum was established based on the high fluorescent intensity in ethanol solution. The components were ultrasonic extracted from Polygonum cuspidatum with 80%vol ethanol at 50℃. Using 1,8-dioxyanthraquinone as standard samples, total anthraquinone content was determined by spectrofluorophotometer at excitation wavelength of 440nm and emission wavelength of 520nm. The detection limit was 0.15μg/ml in the linear range of 0.3μg/ml-16.00μg/ml with the regression equation of y=179.27x+24.498, correlation coefficient (r) of 0.9993, average recovery rate of 99.8% and relative standard deviation (RSD) of 0.87%.
