摘要
目的探讨吡拉西坦氯化钠注射液对大鼠实验性脊髓损伤后水通道蛋白4(Aquaporin 4,AQP4)表达的影响。方法将120只健康SD大鼠随机分为对照组(A组)、手术组(B组)、吡拉西坦组(C组),采用改良Allen法制备脊髓损伤模型,分别在6 h、24 h、72 h和5 d时取大鼠脊髓组织,用免疫组织化学染色法和Western-blot法观察各组AQP4蛋白表达情况。结果与A组相比,B组、C组均从6 h开始出现脊髓水肿,72 h达到高峰,且C组脊髓水肿低于B组。同样AQP4表达在手术后开始升高,72 h后达到高峰,且B组较A组和C组明显增高(P<0.05)。结论 AQP4与脊髓水肿形成有关,吡拉西坦可以抑制脊髓组织内AQP4的基因表达。
Objective To study the effects of piracetam on the expression of AQP4 in rats with experimental spinal cord injury. Methods 120 normal SD rats were randomly divided into control group(group A),surgical operation group(group B) and piracetam group(group C).The rat model of spinal cord injury was established by using improved Allen method.The expression of AQP4 was detected by using Western blotting at 6 h,24 h,72 h and 5 day. Results As compared with group A,spinal cord edema appeared in groups B and C at 6 h after operation,reached the peak at 72 h,and the spinal cord edema in group C was milder than in group B.The AQP4 mRNA and AQP4 expression levels in group B were obviously higher than those in groups A and C(P〈0.05),and they increased at 6 h,reached the peak at 72 h. Conclusion AQP4 may be involved in the formation of spinal cord edema.Piracetam can decrease the expression AQP4 and relieve spinal cord edema.
