摘要 目的探讨阿托伐他汀对脓毒症大鼠血浆一氧化氮(NO)和肝组织匀浆丙二醛(MDA)含量的影响。方法将75只健康雄性大鼠随机分为3组:对照组行假手术;脓毒症组行盲肠结扎穿孔术(CLP);阿托伐他汀组,用40mg/kg阿托伐他汀灌胃,行CLP。分别在术后0h、2h、6h、10h、24h时间点检测血浆NO和肝脏组织匀浆MDA的含量。术后24 h取心脏、肺、肝脏、空肠标本,光镜下检查组织病理学变化。结果脓毒症组、阿托伐他汀组血浆NO的浓度,在术后6h、10h、24h时间点差异有统计学意义(P<0.01)。脓毒症组、阿托伐他汀组肝组织匀浆MDA的含量,在术后2h、6h、10h、24h比较,差异有统计学意义(P<0.05或P<0.01)。光镜下对照组无明显病理改变,脓毒症组有炎细胞浸润,组织充血、坏死、渗出,阿托伐他汀组病理改变介于两者之间。结论阿托伐他汀可能降低脓毒症大鼠模型NO与MDA的含量,减轻炎症反应。 Objective To investigate the influence of atorvastatin on nitric oxide(NO)and malondialdehyde(MDA)of the sepsis rats.Methods A total of 75 male rats were randomized into three groups.The control group was treated with sham-operation.The sepsis group was made with cecal ligation and puncture(CLP).The atorvastatin group was feeded on 40 mg/kg atorvastatin,then the rats were made with CLP.The blood and hepatic tissue samples were separately colleced in postoperative 0 h,2 h,6 h,10 h,24 h in three groups,and the levels of NO and MDA were measured through the spectrophotometera.The specimens of the heart,lung,liver and jejunum of rats were dyed and the tissue pathology was observed under light microscope at 24 h after operation.Results In atorvastatin group,the concentration of NO significantly decreased(P0.01)in comparison to the sepsis group after 6 h,10 h and 24 h,and the MDA were also significantly decreased in comparison to the sepsis group(P0.05 or P0.01).There was no pathological change in control group under the light microscope.In sepsis group,the specimens of the heart,lung,liver and jejunum were all immersed by different inflammatory cells,and hyperemia,necrosis and exudate were shown in the specimen tissue.Conclusion Atorvastatin may postpone the rising velocity of NO and MDA in sepsis rats,and reduce inflammatory reaction.
