摘要
目的 探讨外源性的白介素(IL)-10对柯萨奇B3病毒(CVB3)性心肌炎小鼠治疗作用的机制.方法 将84只Balb/c小鼠随机分为正常组、感染组和治疗组.感染组和治疗组用CVB3感染Balb/c小鼠制备病毒性心肌炎小鼠模型,然后治疗组给予重组小鼠(m)IL-10治疗5天,正常组和感染组在同一时间腹腔注射磷酸盐缓冲溶液,观察小鼠发病及死亡情况,在不同时间段,测定小鼠心肌病毒滴定度、病理积分及心肌组织中IL-2与肿瘤坏死因子的表达,并进行比较.结果 3组小鼠的生存率(χ2=10.562,P〈0.01)、不同时期病理积分(F=3.587~5.794,均P〈0.01)及心肌IL-2和肿瘤坏死因子-α的表达均具有显著性差异(均P〈0.01).感染组和治疗组心肌病毒滴度无明显差异(F=8.389,P〉0.05).结论 IL-10不能降低心肌病毒滴度,但能减少心肌组织中IL-2和肿瘤坏死因子-α的表达,对早期病毒性心肌炎有治疗作用.
Objective To investigate the therapeutic effect of interleukin-10 (IL-10) on viral myocarditis (VMC) induced by Coxsackie virus B3 ( CVB3 ). Methods 84 Balb/c infected mice were randomly divided into three groups: normal group, infection group and treatment group. The VMC mouse models in infection group and treatment group were prepared by inoculating Balb/c mice with CVB3. Then mIL-10 was administered for 5 days in treatment group, and daily phosphate buffered solution (PBS) was injected intraperitoneally in normal group and infection group at the same time. The morbidity and mortality of mice were observed. Virus titer in myocardial tissue, pathological score and the expressions of IL-2 and tumor necrosis factor (TNF) were measured at different periods and made comparison among them. Results There were significant differences in survival rate (χ2 = 10. 562, P 〈 0.01 ), pathological score of different period ( F = 3. 587- 5.794, P〈0.01) and the expressions of IL-2 and TNF-α among three groups (all P 〈 0.01 ). The difference.in virus titer between infection group and treatment group was not significant (F = 8. 389, P 〉 0.05). Conclusion Although IL-10 cannot reduce the virus titer of the heart, it can decrease the expressions of IL-2 and TNF-α in myocardial tissue, and it has therapeutic effect on VMC at early stage.
